Design Notes

The choice of RBS: The RBS (BBa_B0034) was chosen because it has a high translation initiation rate and is compatible with most bacterial hosts. The codon optimization: The codon usage of the ycnE gene was optimized for E. coli expression using the online tool [Optimiser]. The restriction sites: The ycnE gene was flanked by EcoRI and PstI restriction sites to facilitate cloning into standard plasmid vectors.

This part is derived from the genomic sequence of E. hirae GDIAS-5, a strain isolated from pig manure by a research team from Guangdong.
